Vanderbilt team develops a selective TAOK-1 inhibitor and a pan-TAOK activator, expanding tools to investigate the disease

On August 3, 2026, researchers announced the creation of two chemical compounds that could broaden the study of Alzheimer's disease. The molecules were developed at the Vanderbilt University Warren Center for Neuroscience Drug Discovery.

Compounds as research tools

The so-called “tool compounds” interact with specific proteins, modulating their activity up or down. Although most of these compounds are not suitable for use as medicines due to possible side effects, they are essential for understanding the role of a protein in the organism, an important preliminary step in the search for new treatments.

Selective TAOK-1 inhibitor

The study published in ACS Chemical Neuroscience describes the discovery of VU6083859, the first selective inhibitor of the TAOK-1 protein. This protein is associated with Alzheimer's disease, but its function was still poorly understood due to the lack of molecules that allowed its controlled manipulation.

Unexpected pan-TAOK activator

A second compound, VU6080195, displayed an unexpected behavior by activating the three members of the TAOK family instead of inhibiting them. The researchers highlighted the excitement of studying the neurological effects of increased activity of these enzymes, since most previous investigations focused on their inhibition.

Impact on studying the TAOK family

The authors hope that the two compounds will encourage more scientists to investigate the TAOK family, which has received little attention in living models. A deeper understanding of the biology of these proteins may improve the chances of finding effective therapeutic strategies for Alzheimer's and other neurological diseases.

Context of the discovery

Most of the work was carried out at WCNDD, a clinical-stage biotech startup affiliated with Vanderbilt, under the direction of Craig Lindsley. The center also belongs to the newly created Vanderbilt Institute for Therapeutic Advances. The research was funded by the William K. Warren Foundation, the Zenobia and Mark Godschalk Alzheimer’s Research Endowment, the Helen H. and Morris D. Hartman Neurological Research Fund, among other institutional support.
